The detailed insights of the effect of theophylline on human skin are poorly understood. Here, we show that theophylline possesses several striking complex skin-protecting properties in human skin by employing the clinically relevant human full-thickness skin ex vivo organ culture model. Our study encourages further examination of theophylline as a topical candidate for anti-aging treatment.

Our current understanding of Monocyte (MO)-derived macrophages (MACs) origin and their differentiation in peripheral tissues is limited to murine studies and the existence of such activities in human skin remains elusive. By applying our established full-thickness skin organ culture, the results provide the first insights that de novo generation of human dermal CD68+ MACs from an expanding pool of resident CD34+ mesenchymal stem cells is possible also in human skin. Therefore, it would be intriguing to examine the MAC pool in skin diseases (dermatoses) and develop therapeutics targeting pathological intradermal MAC differentiation processes.
